Rapamycin is commonly prescribed to prevent rejection in patients who have undergone kidney transplantation. It works by suppressing immune system activity through inhibition of the mTOR pathway, which helps decrease the risk of the body attacking the transplanted organ. In some cases, it may also be used for certain rare lung conditions such as lymphangioleiomyomatosis (LAM).
Rapamycin is taken orally, usually once daily, with or without food, but it should be taken consistently the same way each day. The dosage is individualized based on blood levels, overall health, and transplant protocols. Tablets should be swallowed whole and not crushed or chewed. Always follow your healthcare provider’s dosing instructions carefully.
Before using rapamycin, inform your doctor if you have liver disease, high cholesterol or triglycerides, infections, healing problems, or if you are scheduled for surgery. Rapamycin may increase the risk of infections and slow wound healing. Avoid live vaccines while taking this medication. Regular blood tests are required to monitor drug levels, kidney function, and lipid levels.
Rapamycin is contraindicated in individuals with known hypersensitivity to sirolimus or any formulation components. It should not be used concurrently with strong immunosuppressive regimens that significantly raise the risk of infections unless supervised by a specialist.
Common side effects include mouth sores, headache, nausea, diarrhea, joint pain, high cholesterol, and increased risk of infections. Serious side effects may include delayed wound healing, lung complications, swelling, blood clots, kidney dysfunction, or severe allergic reactions. Seek medical attention if serious symptoms occur.
Rapamycin may interact with medications that affect liver enzymes, including certain antibiotics, antifungals, antivirals, anticonvulsants, and herbal supplements such as St. John’s wort. Grapefruit and grapefruit juice may significantly alter drug levels and should be avoided. Always inform your doctor about all prescription drugs, OTC medications, and supplements you take.
If you miss a dose, take it as soon as you remember unless it is close to the next scheduled dose. In that case, skip the missed dose and continue with regular dosing. Do not double the dose to make up for a missed one.
Symptoms of overdose may include severe infection, tremors, dizziness, stomach discomfort, or abnormal blood test results. Seek immediate medical attention if an overdose is suspected.
Store at room temperature, away from heat, moisture, and direct light. Keep rapamycin in its original container, tightly closed, and out of reach of children and pets.
